Alcohol oxidase (EC 1.1.3.13; AO) is a 600 kDa homo-octameric enzyme, which has eight identical subunits. Each subunit contains a strongly and non-covalently bound flavin adenine nucleotide (FAD) molecule as the prosthetic group. This enzyme has been isolated and charactrized from a variety of yeasts and bacteria, such as Pichia pastoris, Paecilomyces variotii, Pichia methanolica, and Thermoascus aurantiacus. Alcohol oxidase is the key enzyme of methanol metabolism in methylotrophic yeast species. It catalyzes the first step of methanol metabolism. Thus alcohol oxidase is a rapid and efficient catalyst for oxidation of lower primary alcohols to the corresponding aldehydes using a two-step mechanism: first a reductive half-reaction, where the sustrate is oxidized while FAD is reduced to FADH2, followed by an oxidative half-reaction, where the flavin is reoxidized by O2 and H2O2 is produced. This enzyme is classified as a member of the glucose-methanol-choline (GMC) oxidoreductase family, and the systematic name of this enzyme class is alcohol:oxygen oxidoreductase, which is also called ethanol oxidase.
