Enzyme Activity Measurement of Alpha-L-Fucosidase

Alpha-L-fucosidases (EC 3.2.1.51) are exoglycosidases found in organisms ranging from bacteria to fungi, mollusks, ascidians, and mammals. They catalyze the removal of nonreducing terminal L-fucose residues linked via alpha-1,2, alpha-1,3, alpha-1,4, or alpha-1,6 bonds to oligosaccharides and their conjugates. Alpha-L-fucosidases are divided into two distinct glycoside hydrolase families. Alpha-L-fucosidases that catalyze the hydrolysis using a retaining mechanism, which is shared by the well-known glycoside hydrolase family 29 (GH29). While the enzymes that catalyze the hydrolysis using an inverting mechanism belong to the recently identified glycoside hydrolase family 95 (GH95).

Alpha-L-fucosidases are of considerable interest because they are related to diseases and clinical diagnosis. The deficiency of alpha-L-fucosidase activity is involved in the pathological conditions such as inflammation, cancer, and cystic fibrosis. Severe deficiency causes fucosidosis, an autosomal recessive lysosomal storage disease. Fucosidase activity in serum is used in the diagnosis of patients with early colorectal and hepatocellular cancers. These enzymes also attract attentions due to the central role of fucosylated glycoconjugates in many biological events. Fucosylated glycans play a crucial role in many physiological and pathological processes, including immune response, signal transduction, development, and adhesion processes of pathogens. Since a growing number of research is focused on alpha-L-fucosidases, an accurate activity assay is urgently needed to further characterize the enzyme kinetics.
