α-Mannosidase is an acid hydrolase which is located in plant vacuoles and is thought to be involved with the turnover of N-linked glycoproteins. α-Mannosidase has been shown to inhibit the proliferation of B-lymphocytes. α-Mannosidase from Canavalia ensiformis is a tretamer composed of two subunits that each contain two components at 44 and 66 kDa.

Unit Definition
One Unit of a-D-mannosidase activity is defined as the amount of enzyme required to release one μmole of p-nitrophenol (pNP) per min from p-nitrophenyl-a-D-mannopyranoside (2 mM) in MES buffer (100 mM) and CoCl2 (1 mM) at pH 6.0 at the temperatures indicated.

Preparation Instructions
For assay, this enzyme should be diluted in MES buffer (100 mM), pH 6.0 containing 1 mg/ml BSA. Swirl to mix the enzyme immediately prior to use.
