In vitro
As an AMPK activators, A-769662 activates AMPK with EC 50 of 0.8 mM. A-769662 was equally potent in activating the baculovirus expressed alpha-1,beta-1,gamma-1 recombinant isoform of AMPK with EC50 of 0.7 mM. A-769662-induced AMPK activation occurs by reversible binding.
In vivo
Since AMPK activation inhibits ACC activity, there is a decrease in malonyl CoA levels, leading to reduced lipogenesis and increased fatty acid oxidation.
