In vitro
As a analogue of the cytosine nucleoside, 5-azacytidin can inhibit DNA methyltransferase at low doses and cause hypomethylation of DNA. At high doses,5-azacytidin can directly induce apoptosis by inducing cytotoxicity in abnormal hematopoietic cells in the bone marrow.
In vivo
Given two sequential five-day courses of VP-16, 250 mg/m2 for three days, followed by 5-Az, 300 mg/m2 for two days, this combination therapy refractory to anthracyclines and cytarabine indicates a potential role in treatment of patients with ANLL.
