Diisopropyl-fluorophosphatase (DFPase, EC 3.1.8.2) is one of the phosphotriesterases. DFPase is an enzyme that is able to hydrolyze both toxic organic phosphorus containing compounds and dipeptides with C-terminal proline. DFPases are divided into Mazur-type DFPase which is stimulated by Mn2+ and squid-type DFPase which required Ca2+ ions for activity. The squid-type DFPase hydrolyzes diisopropylfluorophosphate (DFP) more rapidly than O-pinacolyl methylphosphonofluoridate (soman), unlike the Mazur-type DFPase, which hydrolyzes soman more rapidly than DFP.

Both DFP and soman are organophosphorus triesters, including sarin, cyclosarin, and tabun. These organophosphorus triesters act as nerve agents, due to their irreversible inhibition of the acetylcholinesterase. DFPase can detoxify such chemical warfare agents via hydrolysis, and therefore they are of increasing interest since they represent a rational alternative for the currently discussed decontamination strategies of chemical warfare agents. Studies on DFPase contribute to resolving the increasingly harsh environment issues, including the drinking water and the indoor air contamination. In addition, DFPase is also being used in medical applications as an antidote or a therapeutic in preventing organophosphorous poisoning. Since organophosphates are synthetically designed compounds, the hydrolysis of nerve agents cannot be the natural function of DFPase. The improvement of enzymatic activity and the broad substrates selectivity are of key studies.
