Glycerol dehydrogenase (EC 1.1.1.6, also known as glycerin dehydrogenase, NAD+-linked glycerol dehydrogenase, glycerol: NAD+ 2-oxidoreductase, GDH, GlDH, and GlyDH) is an enzyme in the oxidoreductase family that utilizes NAD+ to catalyze the oxidation of glycerol to form glycerone (dihydroxyacetone). This enzyme belongs to the family of oxidoreductase and, more specifically, a metal-dependent alcohol dehydrogenase. The enzyme plays an important role in anaerobic glycerol metabolism. It has been isolated from a number of bacteria, including Enterobacter aerogenes, Bacillus megaterium and so on. Glycerol dehydrogenase bears a global structure as a homooctamer, which is composed of eight identical monomer subunits made up of a single polypeptide chain containing 370 amino acids. Each subunit two distinct domains (N-terminal and C-terminal), which form the active site of the enzyme in a shape of a deep cleft between these two domains. This active site consists of one bound metal ion, one NAD+ nicotinamide ring binding site, and a substrate binding site.

EC 1.1.1.6 acts on the CH-OH group of the substrate glycerol, or, the donor, with NAD+ or NADP+ as the acceptor. The measurement of enzyme activity is commonly based on spectrophotometry, which detects the consumption of NAD+ at 260 nm or the generation of NADH at 340 nm.
