Glycoprotein-fucosylgalactoside alpha-N-acetylgalactosaminyltransferase (EC 2.4.1.40) belongs to the class of glycosyltransferases (GT), which synthesize new glycosidic linkages by adding monosaccharide units from activated donor molecules such as UDPGal in a stereospecific and regiospecific manner to target acceptor molecules. EC 2.4.1.40 specifically transports the hexosyl group from UDP-N-acetyl-alpha-D-galactosamine to glycoprotein-alpha-L-fucosyl-(1,2)-D-galactose, releasing the UDP molecule. The catalytic reaction needs no cofactor but the participation of Mn2+ ion.

GT genes are ubiquitous in vertebrates. They have not been found in invertebrates, plantsyeast, or archaea. The GT genes in humans are involved in the generation of individual variations in blood group structures (A, B, AB, or O). EC 2.4.1.40 contributes to the formation of the blood group A antigen from the H-antigen and performs the final glycosylation step in A antigen synthesis. In addition to the lactoseries and neolactoseries of glycosphingolipid biosynthesis, this enzyme also participates in the biosynthesis of glycan structures such as polysaccharides and complex oligosaccharides, which have vital roles in biological structures and processes including cellular interactions, matrix structures, signaling, pathogenesis, and immunity.
