IMP dehydrogenases belongs to the oxidoreductase family. This enzyme converts inosine 5'-phosphate to produce xanthosine 5'-phosphate, using NAD+ as a cofactor. It is also known as:

Inosine monophosphate (IMP) is produced by de novo purine nucleotide biosynthesis. It is the precursor to both adenine and guanine nucleotides, essential materials for DNA and RNA synthesis. The enzyme IMPDH owns a primary position in virtually every organism. It controls the sole biosynthetic route to guanine nucleotides. Therefore IMP dehydrogenase is extremely important due to its biological significance. The reaction catalyzed by IMPDH is the first committed and rate-limiting step in guanine nucleotide biosynthesis. In humans and other mammals, the enzyme is encoded by two IMPDH genes, IMPDH1 and IMPDH2. In general, human IMPDH1 is expressed constitutively at low levels, while IMPDH2 is amplified only during proliferation and transformation. More specifically, rapidly growing cells have a high demand for guanine nucleotides that generally cannot be sustained by salvage pathways. As a result, the synthesis pathway needs to be activated to supply higher-than-regular amounts of guanine nucleotides, which further explains the key role of IMPDH in cancer and viral infection. The enzyme then evidently has emerged as an attractive target for antiparasitic, antiviral, antibacterial, and immunosuppressive therapies. Hence, IMPDHs from different organisms have been biochemically and structurally characterized, hoping to serve their therapeutic value. In fact, recent studies have already suggested that methiopropamine (MPA) and benzamide riboside are two of its inhibitors.
