L-Amino acid oxidase (LAAO) is a flavoenzyme that catalyzes the deamination of L-amino acids to the corresponding α-keto acids with the production of ammonia (NH3) and hydrogen peroxide (H2O2), simultaneously accompanied with the consumption of H2O. This enzyme was first found present in snake venoms, and subsequently found to exist in algae, bacteria, actinomycete, and fungi. L-amino acid oxidases vary between genera and species, and snake venom LAAOs are best characterized. The catalysis process requires FAD as a cofactor. Structure studies showed that the FAD-binding domain is highly conserved in both prokaryotes and eukaryotes. L-amino acid oxidases have a wide range of substrate specificity: a majority of snake venom sourced LAAOs can act on at least a dozen L-amino acids, while LLAO from Rhodococcus opacus can catalyze 20 L-amino acids. Studies also point out that the thermal stability, optimum pH, molecular weight, and specific activity from different sources are greatly different.
