Enzyme Activity Measurement of Mevaldate Reductase (NADPH) Using Spectrophotometric Assays

Creative Enzymes has a remarkable recorde of providing enzyme activity assays with high-quality services. In the last few years, Creative Enzyme has helped thousands of scientists and researchers fulfill fast and high reproducibility activity measurement, including mevaldate reductase (NADPH).

In enzymology, a mevaldate reductase (NADPH) (EC 1.1.1.33) is an enzyme that catalyzes the chemical reaction targeting (R)-mevalonate, and converts it to mevaldate. The reaction uses NADP+ as cofactor which differs from EC 1.1.1.32. In Sus scrofa (pig), using NADH as cofactor giving one-fifth of the rate obtained with NADPH. This enzyme belongs to the family of oxidoreductases. The systematic name of this enzyme class is (R)-mevalonate:NADP+ oxidoreductase. Another name in common use is mevaldate (reduced nicotinamide adenine dinucleotide phosphate) reductase.

Mevaldate reductase was first found related to mevalonate biosynthesis in rat liver. The high activity of this enzyme helped early workers in the field to obtain a high incorporation rate of labelled mevaldic acid into non-saponifiable lipids in rat liver homogenates, which led to the theory that free mevaldate is an intermediate in sterol biosynthesis. Studies on this enzyme shows it is inhibited by barbiturates. Sodium aminobarbitone and thiobarbituric acid are two main inhibitors of this enzyme. The properties of this enzyme is similar to those of other animal tissue aldehyde reductases (EC 1.1.1.2), and it is suggested that mevaldate reductase be reclassified as one of this group. To assay the activity of this enzyme, the best and efficient way is to employ the spectrophotometric assay method. According to the basic principle of spectrophotometric absorption, measurement of mevaldate reductase activity can be carried out by detecting the consumption of NADP+ or the generation of NADPH. Creative Enzymes is one of the few companies providing accurate and rapid mevaldate reductase activity measurement services. The advanced equipment and professional team here are the best choice of your enzyme assay activities.
