The aldo-keto reductases make up a superfamily of NAD(P)-dependent oxidoreductases with broad physiological roles. These enzymes have broad substrate specificity and may have NADPH-dependent aldehyde reductase activity towards 2-carboxybenzaldehyde, 2-nitrobenzaldehyde and pyridine-2-aldehyde, as indicated by in vitro tests. There is mounting evidence showing that human AKR7A2 can function as a succinic semialdehyde (SSA) reductase in brain and may play a role in the synthesis of γ-hydroxybutyrate (GHB). So far, only succinic semialdehyde reductase in mouse has been well studied. This enzyme has a typical AKR7 family local structure, common to all the members of the aldo-keto reductase superfamily. Due to the specific catalytic chemistry and their promiscuity of the substrate, these enzymes have huge potentials in food processing, fine chemicals, and pharmaceutical industries.
