Isocitrate dehydrogenase (ICDH) is an enzyme in the tricarboxylic acid (TCA) cycle that catalyzes the dehydrogenation and the subsequent decarboxylation of isocitrate to α-ketoglutarate and CO2. EC 1.1.1.42 is a NADP+-linked enzyme that is found in both mitochondria and cytoplasm. The enzyme from some species can also use NAD+ but much more slowly. Isocitrate dehydrogenase (NADP+) catalyzes the important step in the tricarboxylic acid (TCA) cycle. IDH1 and IDH2 are two isoforms of isocitrate dehydrogenase (IDH) in human using NADP+ as the cofactor. These two enzymes are of great clinical significance. Specific mutations in IDH1 have been found in several brain tumors including astrocytoma, oligodendroglioma, and glioblastoma multiforme.
