Triacylglycerol lipase is an enzyme with system name triacylglycerol acylhydrolase. This enzyme catalyses the following chemical reaction:triacylglycerol + H2O↔ diacylglycerol + a carboxylate.

One unit will hydrolyze 1.0 microequivalent of fatty acid from a triglyceride in 1 hr at pH 7.4 at 37 °C. (This is equivalent to approx. 10 microliters of CO2 in 30 minutes.)
One unit will hydrolyze 1.0 micromole p-nitrophenyl phosphate per minute at pH 4.8 at 37 °C.
