Notes
The Y289L substitution broadens donor-substrate acceptance to GalNAc and GalNAz.
Protocol
1. Prepare the reaction system with 50 mM Tris-HCl (pH 7.5) and 6 mM MnCl₂.
2. Use a UDP-GalNAz-to-antibody equivalent ratio of 300:1.
3. Set the enzyme-to-antibody addition amount to 5%.
4. Incubate the reaction at 37°C for 48 hours.
5. Transfer the reaction to -25°C to -15°C to stop the reaction, then proceed with sample analysis.
