L-lysine oxidase (EC is a flavoenzyme that catalyzes the irreversible oxidative deamination of L-lysine to α-keto-ε-aminocaproate, concomitant with the production of ammonia and hydrogen peroxide. This enzyme was originally isolated from Trichoderma viride Y 244-2, and later it was also found in Trichoderma harzianum Rifai and some other marine organisms, such as the rockfish Sebastes schlegelii. Most of the L-amino acid oxidases show a wide range of substrate specificity, but not for L-lysine. However, this is not the case for L-lysine oxidase from T. viride, which exhibits a very high substrate specificity for L-lysine. This enzyme is stable over the wide range of pH and temperature, though the prosthetic flavin group could be released from the enzyme under acidic conditions. L-lysine oxidase is a member of the general oxidoreductase family, which catalyzes the CH-OH group of the substrate and uses NAD+ or NADP+ as the cofactor. The systematic name of this enzyme class is L-lysine:oxygen 2-oxidoreductase (deaminating), which is also known as L-lysine α-oxidase or L-lysyl-α-oxidase. L-lysine oxidase from T. viride is a homodimeric protein with an apparent molecular mass of 112 kDa. However, the molecular mechanisms of the enzymatic reaction and unusually high substrate specificity have not been well explored.

L-lysine oxidase from T. viride is useful in the food industry or fermentation process as an enzyme-based amino acid sensor for quantitative measurement of L-lysine concentration. Moreover, the enzyme has a strong antitumor activity and hence it is considered to be a potential anti-cancer agent.
