Immunoassay for Bioanalysis with Nanozymes

Using the peroxidase mimics or the combined oxidases and peroxidase mimics, then combining with unique physicochemical properties and catalytic activities of nanozymes, we developed immunoassay for bioanalysis based on nanozymes, to detect cancer markers, pathogens, toxins, functional protein, pesticide residues, antibiotics and other biomarkers.

Nanozyme-Based Colorimetric Immunoassays

  • As a commonly used material in biosensing, Fe3O4 nanozymes exhibit not only good magnetism for facile immunomagnetic separation / enrichment but also favorable peroxidase-like activity to catalyze chromogenic reactions. Thus, Creative Enzymes uses them as a bifunctional platform for colorimetric immunosensing of low-level targets.
  • Compared to regular nanozymes, the catalytic properties of these photosensitive nanozymes can be easily controlled by external light, offering an effective mean to manipulate the chromogenic process. With this consideration, we also use photosensitive nanozymes to manufacture several colorimetric immunosensing platforms for tumor markers and antibiotics.

  • We use these nanozymes to detect tumor cells, alpha fetoprotein (AFP), prostate-specific antigen, Apolipoprotein A1, Ebola virus, parathion, chloramphenicol, carcinoembryonic antigen and others in blood, serum, urine, plasma, etc.
  • The peroxidase substrates TMB and H2O2 are added to trigger a chromogenic reaction, giving a remarkable color change from colorless to deep blue. The color change can be distinguished by naked eyes or quantified by UV-vis.

Nanozyme-Based Chemiluminescent Immunoassays

  • By using some peroxidase mimics to replace the natural enzyme, nanozyme-based chemiluminescent immunoassays with better stability have been designed for biosensing.
  • Based on the principle, Creative Enzymes develops a chemiluminescent immunosensor for multiplex detection of cytokines by employing CuS nanozymes as a mimetic catalytic tag. Like HRP, the CuS tag can trigger the chemiluminescence of the luminol-H2O2 system, providing a highly sensitive response for quantification.

Nanozyme-Based Fluorescent Immunoassays

  • Under the catalysis of a nanozyme, some non-fluorescent substrates can be oxidized to products with strong fluorescence. Thus, nanozymes can be employed, instead of natural enzymes, to fabricate fluorescent immunoassays for various analyses.
  • Inspired by these, we design a ratio metric fluorescent immunoassay for C-reactive protein (CRP) based on the oxidase-like activity of MnO2 nanozymes.
