Ribonuclease (RNase) is a very important enzyme in living organisms. The major function of these nucleases is catalyzing the degradation of RNA into smaller components. Ribonucleases can be divided into endoribonucleases and exoribonucleases. Ribonuclease T2 (RNase T2, EC 3.1.27.1) belongs to the endoribonucleases class. It achieves two-stage endonucleolytic cleavage to nucleoside 3'-phosphates and 3'-phosphooligonucleotides with 2',3'-cyclic phosphate intermediates.

RNase T2 family has many members. RNases belonging to the RNase T2 family have attracted little interest because they exhibit unique base preferences with a non-absolute base specificity. However, it is gradually known that RNase T2 family enzymes are ubiquitous from viruses to human cells, resulting in a great deal of attention paid to the study of protein structure and enzymatic properties. Nowadays, one of the best-characterized enzymes is RNase Rh isolated from Rhizopus niveus. In addition, members of RNase T2 family include RNase MC1 from bitter gourd seeds, RNase NW and RNase NGR3 from Nicotiana glutinosa, RNase LE and LX from tomato, RNase LC1 and RNase LC2 from sponge gourd seeds, RNase S2 from tobacco, RNase T2 from A. oryzae, and RNase M from A. saito. The discovery of new enzymes from different sources enriches the diversity, and accurate activity assays are efficient approaches for enzyme characterization.
