Enzyme Activity Measurement of Cellulose 1,4-Beta-Cellobiosidase (Reducing End)

Enzyme assay services of Creative Enzymes stand out with high quality and rapid turnovers. Combining the cutting-edge equipment and the advanced techniques, Creative Enzymes has presented the art of enzyme activity assays in satisfying both industrial and research needs. We are specialized in testing hydrolases such as cellulose 1,4-beta-cellobiosidase (reducing end).

Cellulose 1,4-beta-cellobiosidase (reducing end) (Cellobiohydrolase, EC 3.2.1.176) hydrolyzes a β-1,4-glycosidic bond from the reducing end of cellulose and releases cellobiose as the major product. The degradation of polysaccharides by hydrolysis is a biological process with promising industrial applications, although this process needs three other cellulases besides cellobiohydrolases. These specialized enzymes may be secreted independently or, in some organisms, are involved in the formation of the cellulosome, a protein complex comprising an array of hydrolytic enzymes that act in concert to efficiently degrade cellulose.

Cellobiohydrolases have been found in glycoside hydrolase families 7 (GH7) and 48 (GH48). GH7 cellobiohydrolase enzymes exhibit a jelly-roll β-sandwich fold structure and utilize two catalytic glutamates in a retaining hydrolytic mechanism. The GH48 family are predominately classified as (α/α)6-fold enzymes that may utilize aspartate and glutamate as base and acid donors, respectively, for substrate cleavage and release of cellobiose as the main product through an inverting mechanism. Considering the applications in biocatalysis, chemical synthesis, and environmental chemist, more and more variants of this enzyme are being developed, including creating a diverse mutant library and optimizing the composition of enzyme mixtures to hydrolyze the biomass. The activity measurement of cellobiolydrolases is then in a growing demand. The assay is usually performed by detecting the hydrolysis products (cellobiose or glucose) in the supernatant through HPLC. Creative Enzymes’ best-performed chromatography can achieve both rapid and accurate outputs for various tests.
