Enzyme Activity Measurement of Glycoprotein 3-Alpha-L-Fucosyltransferase Using Spectrophotometric Assays

Glycoprotein-3-alpha-L-fucosyltransferase belongs to the hexosyltransferases, more specifically, this enzyme transfers L-fucosy group to N-linked oligosaccharide structures (N-glycans). This enzyme has specificity for N-glycans with one unsubstituted non-reducing terminal GlcNAc residue. The catalytic reaction is similar to that of glycoprotein 6-alpha-L-fucosyltransferase which also transfers L-fucosy group from GDP-beta-L-fucose, but forming alpha 1,6-linkage rather than alpha 1,3-linkage. The enzymatic reaction needs the participation of Mn2+ ion.

Glycoprotein-3-alpha-L-fucosyltransferase takes part in the various types of N-glycan biosynthesis pathways. Therefore, the products of this enzyme are widely distributed from plants to insects and some other invertebrates. This enzyme has great potentials in drug development and pharmacology. Studies have shown that the RNAi lines, generated by RNA interference technology, obtaining a targeted down-regulation FucT gene, which providing a robust tool for the production of therapeutically relevant glycoproteins in plants with a humanized N -glycan structure. As for the treatment of diseases, this enzyme is a target for the rational inhibitor design for medication in the treatment of rheumatoid arthritis. Although the crystal structure of this enzyme has not been identified yet, some modifications including mutations are already tested.
