Enzymes Expression in Other Yeasts

In addition to the most common expression system such as Saccharomyces cerevisiae and Pichia pastoris, several other yeast expression systems are under development. The most popular yeast stains include Arxula adeninivorans, Kluyveromyces lactis, Yarrowia lipolytica, and Schizosaccharomyces pombe showing unique advantages that the typical expression systems do not offer. A. adeninivorans seems an attractive host for heterologous gene expression due to competitive characteristics such as thermos-tolerance and halo-tolerance. As an expression host, Kluyveromyces lactis is best known for its use in commercial production of the milk-clotting enzyme bovine chymosin. K. lactis has several advantages over other yeast expression systems: easy genetic manipulation, the ability to use both integrative and episomal expression vectors, and the availability of a fully sequenced genome information. Compared to the methylotrophic yeasts, K. lactis is able to grow in standard yeast medium so the fermentation process is safer. In addition, enzymes from K. lactis have GRAS (generally regarded as safe) FDA status, which permits wide use in various food, nutrition, and feed applications.

Due to the approvement for several GRAS industrial processes by FDA, Yarrowia lipolytica has also been demonstrated to be a good alternative expression system. It presents numerous advantages as a heterologous protein expression system. Firstly, it has the inherent ability to secrete high molecular weight enzymes. Secondly, the co-translational translocation pathway contributes forming enzymes with the correct structures and functions. Thirdly, the genome is completely sequenced and annotated. Combined with the well-studied molecular biology, the whole construction process is easy to accomplish.

Schizosaccharomyces pombe is considered as an attractive host for the expression of proteins with complicated structures, such as glycosylated proteins derived from higher animals. Several proteins are reported successfully expressed in Sc. pombe. Sc. pombe has also been used as a host for production of enzymes to mediate biotransformations. Since Sc. pombe expression system is not fully developed yet, many possibilities attract a number of ongoing research on the yeast as a promising expression system.
