Alkaline phosphatase (ALP, EC 3.1.3.1) is a nonspecific phosphomonoesterase that catalyzes the hydrolysis reaction called dephosphorylation. This enzyme acts on many types of molecules, such as nucleotides, proteins, and alkaloids, and removes phosphate groups from them. As the name suggests, alkaline phosphatases are most effective in an alkaline environment. It is produced by most organisms and there is considerable homology among species from bacteria to humans.

Alkaline phosphatases from different species have been well studied. One of the most studied is the enzyme from E. coli. A variant of E. coli alkaline phosphatase was generated with improved activity and thermostability compared to the wild type enzyme. This enzyme has become a useful tool in molecular biology experiments for removing phosphate monoesters to prevent self-ligation, which is undesirable during plasmid DNA cloning. In the food industry, quantification of alkaline phosphatase is used for determining the treatment temperature for milk pasteurization. In clinical treatment, the determination of ALP is mainly used for diagnosis of bone and hepatobiliary system diseases, in particular the differential diagnosis of jaundice.
