Case 1: Discovery of Two Novel Oxidases Using a High-Throughput Activity Screen

The discovery of new enzymes is both difficult and essential for biotechnology. Using a high-throughput screening approach, researchers tested 96 putative oxidases against 23 substrates and identified two novel enzymes. The first, N-acetyl-D-hexosamine oxidase (EC 1.1.3.29) from Ralstonia solanacearum, is a vanillyl alcohol oxidase-like flavoprotein active on N-acetylglucosamine and N-acetylgalactosamine; prior to this work, its activity was considered orphaned (known function without sequence). The second, a long-chain alcohol oxidase (EC 1.1.3.20) from an uncultured marine Euryarchaeota, is active on various fatty alcohols (preferentially 1-dodecanol) and shows no sequence similarity to known LCAOs, making it a novel representative of this enzyme class.

Case 2: Effects of Simulated Digestion on AABP-2B

This study examined the impact of simulated saliva–gastrointestinal digestion on Anemarrhena asphodeloides polysaccharide B (AABP-2B), focusing on its structural integrity, α-glucosidase inhibition, and influence on gut microbiota. Results showed that AABP-2B maintained its molecular weight and released no free monosaccharides, indicating resistance to degradation. Its inhibitory activity against α-glucosidase also remained intact, likely due to preserved structural features. In vitro fecal fermentation demonstrated that digested AABP-2B reshaped the gut microbiota, increasing beneficial genera such as Prevotella, Faecalibacterium, and Megasphaera, while suppressing pathogens. Additionally, short-chain fatty acid production increased significantly. These findings highlight AABP-2B's potential as a prebiotic or functional food for gut health.
