Lipoprotein lipase (LPL, EC 3.1.1.34) is first synthesized in the tissues and then transported to the surface of endothelial cells of the capillary beds where it presents the natural functions. LPL hydrolyzes chylomicrons and very low-density lipoproteins to release free fatty acids (FFAs), generates remnant lipoproteins, and gives rise to the formation of high-density lipoproteins (HDL). For example, the activity deficiency causes hypertriglyceridemia, whereas overexpression of LPL may increase insulin sensitivity and promote obesity. Although the crystal structure has not been identified yet, this enzyme is always the hotspot on the researches.
