Blood-group-substance endo-1,4-beta-galactosidase (EABase, EC 3.2.1.102) is the first discovered member of glycoside hydrolase family GH98, denoting its endoglycosidase activity on both A-antigen and B-antigen. More specifically, EABase catalyzes the hydrolysis the 1,4-beta-D-galactosyl linkages adjacent to 1,3-alpha-D-galactosyl or N-acetylgalactosaminyl residues, and a 1,2-alpha-D-fucosyl residue. EABase completely removes the whole trisaccharide of the A and B antigen from glycans on the host cell surface, leaving the antigen-null red blood cells. The main antigenic differences in AOB group system is that the consistence of trisaccharide moiety attached to glycoproteins and lipids on the surface of red blood cells. The trisaccharide of the A-antigen (GalNAcα-1,3-(Fucα-1,2)-Galβ-) terminates with an N-acetylgalactosamine, whereas, in the B-antigen (Galα-1,3-(Fucα-1,2)- Galβ-), the terminal sugar is galactose. Glycans of O-type cells lacking this terminal sugar thus present the core α-Fuc-1,2-ß-Gal- structure and are nonantigenic for the vast majority of individuals. Enzymatic removal of the terminal N-acetylgalactosamine or galactose of A- or B- antigens, respectively, to generate O-type blood has long been proposed as a means of converting erythrocytes into a universal and antigen-null donor type, which has significance in immune response research. Activity measurement for this enzyme is usually performed by fluorometric assays.
