Arachidonate 5-lipoxygenase (EC is a nonheme, iron-containing enzyme which belongs to the lipoxygenase family. This enzyme is involved in the biosynthesis of leukotrienes (LTs) by a two-step catalytic reaction. Arachidonate 5-lipoxygenase (5-LO) first catalyzes oxygenation of arachidonic acid (AA) to 5(S)-hydroperoxy-6-trans-8,11,14-cis-eicosatetraenoic acid (5-HPETE), which further undergoes dehydration to produce the allylic epoxide leukotriene A4.

In humans, this enzyme is of high research values due to its vital position in essential fatty acid (EFA) metabolism and the potential clinical significances. Leukotrienes, the product of this enzyme-catalyzed reaction, is used in lipid signaling to convey information for the regulation of immune responses. In drug and medicine development, this enzyme has become a research target for many diseases. For inflammation and allergy, studies implicate that arachidonate 5-lipoxygenase contributes to both innate immunity and the development and progression of excessive and chronic inflammatory responses. For hypersensitivity reactions, this enzyme leads to non-allergic reactions of the respiratory system and skin, or contributes to hypersensitivity responses of the respiratory system to cold air and alcohol beverages. Discovery of the inhibitors of arachidonate 5-lipoxygenase is an effective searching means of clinical meaningful drugs, such as one of successful inhibitors with the medical name Zileuton which is used for the maintenance treatment of asthma.
